To discover bioactive small molecules ― bioprobes, we have constructed the in-house phenotypic evaluation (iHOPE) system where the biological activities, such as cytotoxicity, differentiation, morphology, of test samples against several cancer cell lines, bacteria, pathogenic fungi, and a malaria parasite, can be exhaustively evaluated.
Here, we examine anti-cancer, anti-microbial, anti-malaria activity of a sample by using following cells and assay methods. All assays are carried out in 384-well format.
The human cervix epidermoid carcinoma cell line HeLa and the human promyelocytic leukemia cell line HL-60 are used in the assay. Anti-proliferative activity of a sample against these cancer cells is evaluated by using WST-8 assay.
The following microorganisms are used as test strains in the assay; Staphylococcus aureus 209, Escherichia coli HO141, Candida albicans JCM1542, Aspergillus fumigatus Af293, and Pyricularia oryzae kita-1. Antimicrobial activity tests are carried out by a standard microdilution method.
Plasmodium falciparum 3D7 is used as a test strain in the assay. Anti-malarial activity of a test sample is evaluated by using pLDH assay.
Not only pure compounds, but also microbial/plant extracts can be tested. Please prepare the sufficient amount of sample for the assay by referring the following table. Note that samples should be dissolved in DMSO.
